Route E21B now provides daily service to Kowloon from morning to noon and to Tung Chung from afternoon to evening, while the rest of the day is served by Routes E21 and E21A.
With the continuous development of Tung Chung West and Tung Chung North and the increasing population, the existing Route E21A serving between Yat Tung Estate in Tung Chung West and Oi Man has to pass through Tung Chung Town Centre and Tung Chung North before entering North Lantau Highway, which is quite time-consuming for the residents of Tung Chung West. In view of this, Citybus proposed in the 2021-2022 Bus Route Planning Programme that one special morning departure of Route E21A from Mun Tung Estate be operated from Mondays to Fridays, and that after Shun Tung Road (westbound), the route would be diverted to Tat Tung Road, Shun Tung Road (eastbound), Yu Tung Road, and then return to its original routing on the North Lantau Highway, no longer detouring via Tung Chung North, in order to provide residents of Tung Chung West with a special express service to the urban areas of Kowloon. At the same time, one trip of Route E21 between Mondays and Fridays to the urban area would be diverted to Tung Chung North instead of Tung Chung Town Centre, and both changes were implemented from 23 August 2021.[1][2] As this mode of operation would reduce the travelling time from Tung Chung West to the urban area, the Islands District Councillor (Yat Tung Estate North Constituency), Mr. Fong Lung-fei (方龍飛), has repeatedly discussed with Citybus and the Transport Department (TD) to expand the time slots of this mode of operation.
As a result of the efforts of Tung Chung residents, Citybus and the Transport Department proposed in the 2022-2023 Bus Route Planning Programme of the following year to extend the morning special departure of Route E21A to all urban-bound trips from morning to noon, and to change the afternoon to last Tung Chung-bound trips of Route E21A to Tung Chung Town Centre after the North Lantau Highway, and then to Mun Tung Estate and Yat Tung Estate, both of which would be separately numbered as E21B. To tie in with the change, the routing of the morning special departure of Route E21 would also been extended to cover all urban-bound trips from the morning to midday, and the afternoon to last Airport-bound trips of Route E21 would been diverted to run via Tung Chung North instead of Tung Chung Town Centre, both of which are separately numbered as E21D.

Outside service hours of this route, passengers may use the following routes between Lantau and Sham Shui Po/Mong Kok:
Half-fare for children aged below 12 and senior citizens aged 65 or above; the remainder of the fare after half-fare will be counted as 10 cents.
Elderly persons aged 65 or above using Senior Octopus Cards or Personalised Octopus cards, Hong Kong residents aged 60 to 64 using JoyYou Card, and eligible persons with disabilities using Personalised Octopus cards with "Persons with Disabilities Status" can enjoy a flat fare of $2.0 per journey under the $2 Concessionary Fare. If the concessionary fare for eligible persons with disabilities aged below 12 and senior citizens aged 65 or above is less than $2.0 after a 50% discount from the regular fare, the beneficiary is only required to pay the original concessionary fare; if the original full fare for Hong Kong residents aged 60 to 64 or eligible persons with disabilities is already less than $2.0, the beneficiary is also only required to pay the original full fare.
Passengers are required to tender exact fare by cash or by Octopus card when boarding. No exact change will be given.
Passengers may also use AlipayHK "EasyGo", Alipay, QuickPass, BoC Pay "RideCode", WeChat Pay HK or WeChat "Ride Code", Visa, Mastercard and UnionPay cards with inductive payment capabilities, as well as Apple Pay, Google Pay, Samsung Pay and Huawei Pay mobile payment platforms for fare payment. Passengers using the above payment method can only enjoy the interchange concessions with other Citybus solely-operated routes or the Citybus departures of the jointly-operated routes, and cannot enjoy the "Public Transport Fare Subsidy Scheme" nor the "Public Transport Fare Concession Scheme for the Elderly and Eligible Persons with Disabilities".
*Two-way section fare - Passengers are required to pay by Octopus card or mobile e-payment platforms, and to confirm once with the same card or same account when getting on and off the bus. If other means of payment is used or the card is not reconfirmed on alighing, the fare from the boarding stop to the final destination will be charged.

At present, this route and Route E21B are served by a total of 9 double-decker buses, with Enviro500 MMC 12m normal version (8507-8524) and 12.8m (6501-6588) equipped with luggage racks, and Enviro500 MMC 12m luxury version (80XX) and 12.8m (68XX, 638XX) of the Cityflyer fleet are used on some of the trips.
